Union leader Cord McNally is protecting a routine gold shipment when his troops are attacked by Confederate forces. Not only does he lose the gold, but one of his strongest officers is killed in the raid. At the end of the Civil War, McNally learns that the raiders had help from the inside, and he vows to uncover the two traitors. After a chance encounter with one of the turncoats, McNally travels to the town of Rio Lobo and makes an unexpected discovery.

When the stranger is assassinated in their presence, the vacation turns dangerous. Before dying, the stranger passes along a secret to Jill. Then, to keep the couple silent, a band of foreign assassins kidnaps their daughter. Offered no help by the police, Bob and Jill hunt for their daughter as they try to understand the information that they have.

Shortly after circus owner Sue Farnum (Dale Evans), her assistant, Gabby (George "Gabby" Hayes), and her late father's ex-partner, Slim Phillips (Addison Richards), accidentally interrupt the filming of a movie starring singing cowboy Roy Rogers (Roy Rogers), Slim is kidnapped. Sue suspects that William Ripley (Grant Withers), a crooked businessman who's trying to steal her circus, is behind the kidnapping. While Sue and Gabby set up a special performance, Roy tracks Slim's kidnappers.
