After Sid (John Leguizamo) pilfers some dinosaur eggs, Manny (Ray Romano), Diego (Denis Leary) and the rest of their prehistoric pals enter a mysterious, underground world to rescue the hapless sloth. During the course of their mission, the gang encounters many creatures great and small, including hungry carnivores and a relentless, dinosaur-hunting weasel named Buck.

Manny the mammoth starts to worry when his daughter Peaches announces her engagement. Still unlucky in love, Sid the sloth volunteers to plan the couple's wedding. To Manny's dismay, nothing can stop the upcoming nuptials, except maybe the end of the world. When Scrat accidentally launches himself into outer space, he sets off a chain reaction that sends an asteroid hurtling toward Earth. Once the night guard at the Museum of Natural History, Larry Daley (Ben Stiller) is now a successful purveyor of infomercial products. Soon after learning that his museum friends have been shipped to the Smithsonian Institution, Larry receives a distress call from Jedediah, who warns him that four of history's greatest villains have come to life and are planning to take over the Smithsonian first, then the world. Larry joins forces with friends old and new to stop the quartet's nefarious plans.

When the exhibits at New York's Natural History Museum start behaving strangely, Larry Daley (Ben Stiller) -- now the director of nighttime operations -- must find out the cause. He learns that the Tablet, which magically brings Teddy Roosevelt (Robin Williams), Jedediah (Owen Wilson) and the other exhibits to life at night, has started to decay.
